COMBINATION FORMULATIONS:
B-Complex Syrup (B1 HCl 5 mg, B2 2.5 mg, B6 HCl 1.5 mg, B12 5 mcg, Niacinamide 50mg, D-Panthenol 5 mg per 15ml)
PHYSIOLOGICAL ROLE
Pyroxidine and pyrodoxamine are readily oxidized to pyridoxal phosphate the coenzyme form. Pyroxidal dependent enzymes include transaminase and decarboxylases involved in synthesis of non essential aminoacid, tryptophan and sulphur containing aminoacid metabolism, formation, of 5-HT, dopamine, histamine, GABA and aminolevulinic acid.
PHARMCOKINETICS
All the three forms are well absorbed from intestine. They are oxidized in the body and excreted as pyrodoxic acid.
DRUG INTERACTIONS
- Isoniazid produces a pyridoxine deficiency state.
- Hydralazine, cycloserine and penicillamine also interfere with pyridoxine utilization and action.
- Oral contraceptives reduce pyridoxal phosphate level.
- Pyridoxine reduces dopamine availability in brain, abolishing the therapeutic effect in Parkinsonism.
- 4- Deoxypyrodoxine is a vita.B6 antagonist.
THERAPEUTIC USES
- Prophylactically (2-5mg daily) in infants with deficiency of other B vitamins.
- Used in Acute Isoniazid poisoning.
- To treat mental symptoms in women on oral contraceptives (50 mg daily).
- In anemia and homocystinuria.
- Convulsions in infants and children.
