MECHANISM OF ACTION:
The Analgesic action is due to rising pain threshold but has weak peripheral anti-inflammatory component and prevention of PG mediated sensitization of nerve endings.
Antipyretic action is due to resetting of hypothalamic thermostat and rapidly reduces fever by promoting heat loss.
Anti-inflammatory action is due to inhibition of Prostaglandin synthesis.
PHARMACOKINETICS: Its well absorbed orally; 1/3rd is protein bound in plasma and it is uniformly distributed in body. Plasma t1/2 is 2-3 hrs. Effect lasts for 3-5 hrs.
ADVERSE EFFECTS: Well tolerated drug but nausea and rashes occur occasionally. Leukopenia is rare.
USES: Analgesic, antipyretic, anti-inflammatory
MECHANISM OF ACTION OF CAFFEINE:
CNS stimulant by inhibiting cyclic nucleotide phosphodiesterase thus preventing the conversion of cyclic AMP to 5’ AMP.
ADVERSE EFFECT OF CAFFEINE: It may produce confusion, tremor, insomnia, excitement, mild delirium.
USES OF CAFFEINE: As a stimulant, headache reliever, bronchial asthma, diuretic.
