MECHANISM OF ACTION:
By accumulating in acidic vessels of parasite and because of its weakly basic nature it raises vesicular pH and thereby interferes with degradation of hemoglobin by parasitic lysosome.
PHARMACOKINETICS:
About 50% gets bound in plasma. It has high affinity for melanin and nuclear chromatin gets tightly bound to these constituent and is concentrated in liver, spleen, kidney, lungs, skin, leucocytes etc. Plasma t1/2 is 3-10 days. Partly metabolized by liver and excreted in urine.
ADVERSE EFFECTS:
Nausea, vomiting, anorexia, itching, epigastric pain, uneasiness, headache.
